マイクロアレイ法を用いたTh1,Th2型免疫応答の分類化と免疫疾患の病態解析

研究概要	Th1またはTh2主導の免疫応答が各種免疫疾患の異なる病態を引き起こしていると考えられている。我々はマウスのTh1、Th2細胞及び免疫疾患モデルを用いてTh1あるいはTh2主導の免疫応答における典型的な遺伝子発現パターンを明らかにし、ヒトの免疫疾患の病態解析に応用しようと考えた。まずはじめにT細胞レセプタートランスジェニックマウス(DO11.10)の脾細胞から純粋なTh1、Th2細胞を調整し、そのRNAを用いてTh1、Th2細胞に特異的な遺伝子発現を、DNAマイクロアレイ法を用いて調べた。この結果、約8700個の遺伝子のうちTh1に特異的に発現するものとして82個、Th2に特異的なものとして52個の遺伝子が同定された。このほか、Representational Difference Analysis(RDA法)においても19個の遺伝子がTh1あるいはTh2に特異的な遺伝子として同定された。これらの解析結果をもとに約150の遺伝子を選択して、RT-PCRにより平均500bpの長さのcDNA断片をそれぞれクローン化した。これを用いてTh1、Th2細胞に特異的な遺伝子発現を検出するための独自のDNAアレイフィルターを作製した。次に、マウスのTh1、Th2細胞から抽出したRNAを用いて、このアレイフィルターがTh1/Th2細胞に特異的な遺伝子発現を検出するのに有効であることを確認した。現在、マウスの免疫疾患モデルのリンパ球を材料にして、このDNAアレイフィルターを用いて生体内におけるTh1型あるいはTh2型免疫応答の類型化作業を行っている。平成12年度中にはマウスの結果をふまえて、改良したヒト用のフィルターを作製し、免疫疾患患者の臨床材料を用いた病態解析を進めていく。
