SNPの数理解析とSNP利用研究のための統計解析手法の開発

研究概要	(1)SNPマーカーを用いたgenomewide screeningおよびcandidate gene approachにおける統計学上の問題点を分析し、common diseaseの感受性遺伝子を効率的に探索するためのガイドラインの作成(マーカーの選択や配置も含む)を目指して研究を行なった。種々のアプローチ(ケース・コントロール研究、TDT、罹患同胞対法)に対して、ゲノムワイドにSNPマーカーを設定して行なう疾患感受性遺伝子探索に必要なサンプルサイズを計算した。その結果、マーカーSNPと感受性遺伝子との間に連鎖不平衡が存在していても、両者のアリル頻度が異なると、感受性遺伝子を検出することが著しく困難となることが確認された。感受性遺伝子のアリル頻度は常に未知であることから、連鎖不平衡が保たれていると考えられる範囲内に、異なるアリル頻度を示す複数のSNPを設定しない限り効率よく感受性遺伝子を検出することができないことが確認された。しかし、SNPマーカーの数を増やせば、それに伴い第1種の過誤が起こる確率が増加してしまうことから、genomewide screeningにおいては、SNPマーカー中に真の感受性SNPが含まれていない場合には、感受性遺伝子の検出は困難であることが明らかとなった。現在は、マーカーとして用いるSNPの頻度分布や集団の連鎖不平衡を考慮して、感受性遺伝子同定に必要なSNPマーカー数を決定するガイドラインの作成を行なっている。 (2)集団中の連鎖不平衡の理論予測を行なうために、2遺伝子座無限対立遺伝子モデルのシミュレーションプログラムの構築を行なった。現在、連鎖不平衡の解析解を求めるべく、2遺伝子座を同時に扱う拡散方程式および遺伝子合体理論の研究を行なっている。 (3)日本人、タイ人、キルギス人集団に対して、複数の遺伝子中のSNPを解析した。HLA領域(6p21.3)を主な解析対象としたが、これ以外の領域についても今後解析を行なう。遺伝子解析終了後、連鎖不平衡の程度を異なる染色体領域について推定し、理論予測との比較を行なう。
