TNFにより誘導される遺伝子群の同定とその治療への応用

研究概要	<背景と目的> TNFは慢性関節リウマチや、グラム陰性悍菌感染時にみられるエンドトキシンショックなどにおいて中心的な役割を果たすことが知られており、またある種の腫瘍細胞にはアポトーシスを誘導することが知られている。しかしながら、大多数の他の細胞にはアポトーシスを誘導せず、むしろ炎症性サイトカインの産生や接着因子などの発現上昇を誘導することから、TNF刺激時にはアポトーシスを誘導するシグナル伝達経路と同時に抗アポトーシス遺伝子の誘導がもたらされると考えられている。しかしながら、このような多彩な機能を有するTNFにより誘導される遺伝子群については完全には明かにされていない。我々の作製したTNF receptor-associated factor(TRAF)5/TRAF2のダブルノックアウトマウス(DKO)由来の胎児線維芽細胞(MEF)はTNF刺激によるNF-κBの活性化の障害とTNF誘導性細胞死に対する感受性の亢進が認められる。そこでcDNAマイクロアレイを用い、野性型およびDKO由来のMEF細胞をTNF刺激することにより誘導される遺伝子群の発現パターンを検討し、DKO MEFで特異的に発現の低下した遺伝子群をcDNAマイクロアレイを用いて同定することを目的とする。 <検討結果> <考察> 今後の方針として発現誘導に差の確認された未知の遺伝子については、全長のシークエンスを行うこと、およびそのcDNAをレトロウイルスベクターに組み込み、DKO MEFに遺伝子導入し、TNF誘導性細胞死に対して抵抗性となるかを検討する予定である。これらの解析を通じて多彩な機能を有するTNFにより誘導される遺伝子群の全貌が明らかとなることが期待される。
