放射線抵抗細菌Deinococcus radioduransのDNA2本鎖切断修復

研究概要	真性綿菌D.radioduransは放射線超耐性の生物である。本研究は、放射線超耐性の仕組みを明らかにすることである。1)分離してある放射線感受性変異の原因遺伝子をクローニングするために、λファージのコスミドライブラリーを用いて形質転換し、放射線抵抗性を示すλファージを検索し、recA、recN、lexAなどのhomologをクローニングした。このアプローチではうまくクローニングできないものに関しては、大腸菌とD.radioduransとのシャトルベクターを用いてクローニングを継続中である。2)1999年に発表された全ゲノム配列に含まれる機能未知のORFは約1000個である。この中に放射線超耐性を決定する遺伝子があると考え、網羅的なゲノム破壊を計画した。そのために先ず、PCRによって増幅した全長遺伝子の中程にクロラムフェニコール遺伝子を挿入し、double crossing overによって染色体遺伝子破壊を試みた。この方法によってrecA遺伝子やlexA遺伝子を破壊することができたので、未知の遺伝子に対しても同様に破壊を行っている。また、未知のORFの中程をPCR増幅しクローニングした断片を、プラスミドに乗せたままでsingle crossing overによって染色体に挿入することでも変異体が得られることがわかった。今後はこれらの方法を用いて1000個のORFを網羅的に破壊し、放射線耐性に関わる遺伝子を突き止めてゆきたい。3)DNA損傷をシグナルとして発現する遺伝子を検索するために、シャトルベクターにプロモーターの無い大腸菌lacZ遺伝子をクローニングした。この5'上流に、D.radioduransの染色体断片をクローニングし、X-galで青いコロニーを作るものを集めて解析することのできるpromoter trapを作成した。これを用いて今後は放射線誘導遺伝子の解析を行う予定である。
