研究概要	膜タンパク質は、ゲノムから得られる全タンパク質の中で非常に重要な位置を占めている。第1に、細胞内外の情報伝達、エネルギー変換、物質の輸送を行うタンパク質はほとんど膜タンパク質であり、生理的に非常に重要な生物の部品となっている。第2に、薬のターゲットの多くは受容体(細胞膜内にある膜タンパク質)と考えられており、膜タンパク質に関する情報は製薬関係を中心に産業的にも重要である。第3に、ゲノム情報から得られるアミノ酸配列の中には多くの未知配列があるが、そのかなりの割合が膜タンパク質と予測されている。そこで本研究では、アミノ酸配列から膜タンパク質の構造・機能を推定するための基礎研究と解析ツールの開発を目的とし、以下のことを行うこととした。(1)各生物種の持つすべてのアミノ酸配列情報が与えられるゲノム時代に入ったことから、大量のアミノ酸配列を高速かつ高精度に解析できるようにする。(2)受容体タンパク質を中心とした膜タンパク質と基質のドッキングの問題が解けると、コンピュータ内で自由に薬のデザインが可能になると考えられる。そうした(製薬)産業からの期待にも答えられるような研究を行う。美宅は本研究グループのとりまとめを行うと同時に、大量のアミノ酸配列からできるだけ多くの構造・機能情報(シグナルペプチドの予測、膜タンパク質の判別、膜貫通セグメントの予測、その配置予測など)を自動的に与えるような解析ツールを開発した。園山は、膜タンパク質解析ツールを用いて、次々と得られつつある全ゲノム解析が完了した生物に対して情報処理を行い、膜タンパク質の分布や生物ごとのタンパク質の構成について検討した。倭は、特にイオンポンプであるバクテリオロドプシンや受容体として初めて立体構造が明らかになったロドプシンなどについて、機能発現機構を分子動力学シミュレーションを用いて明らかにした。山登は、チャネルタンパク質のイオン認識・透過機構を、計算機シミュレーションを用いて原子レベルで解明するために、計算機シミュレーションによって基質の結合・透過など機能の全過程の追跡を試みた。
