研究概要	Musashi1(Msi1)は哺乳類初期発生において脳室周辺部の中枢神経系未分化幹細胞において強く発現しているRNA結合蛋白質である。本研究ではm遺伝子産物の神経幹細胞の分化制御における役割を検討した。 1.Msi1は、RNA結合蛋白質であり、そのin vivoで標的となっているRNAの同定を行った。Msi1は、in vivoでNotch-シグナル細胞内アンタゴニストをコードするm-Numb mRNAの3'-UTRに結合していることが示された。luciferase-mNumb 3'UTRキメラレポター遺伝子由来のLuciferase活性は、外来性のMsi1の導入量依存的に低下することが明らかとなった。しかしながら、luciferase-mNumb 3'UTRキメラレポーター遺伝子由来の転写産物の量は、外来性のMsi1の導入量にかかわらず、一定であった。よってMsi1は、m-Numb mRNAの3'-UTRに結合し、この翻訳を抑制しているものと考えられる。 2.Hes1-luciferase活性を指標に、活性化型NotchとともにNIH3T3に導入した外来性Msi1によるNotchシグナルへの影響を検討したところ、外来性Msi1はNotchシグナルを活性化させることが明らかとなった。 3.Neurosphere法を用いて、musashi遺伝子活性が減少した時の神経幹細胞の性質について検討したところ、よってmsi1とその関連遺伝子であるmsi2は協調して神経幹細胞の増殖あるいは生存維持に必須な役割を担っていると考えられた。
