研究概要	大脳皮質の神経回路の入力依存的発達にはシナプス長期抑圧が関与していると考えられているが、本年度は、このような長期抑圧が、生理的な状態の大脳に存在するかどうかの解明を試みた。また、大脳神経回路の発達には異所性投射細胞の退行や細胞死を伴うことが知られているが、なぜそのような神経細胞死が起きるかのメカニズム解明及び大脳神経回路の形成にはシナプス機能の発達が不可欠と思われるがそのプロセスにおけるシナプス受容体機能の発達メカニズムの解明をもめざした。その結果、以下の知見を得た。 1)大脳皮質スライス標本で長期抑圧を起こすことが知られている求心路の1Hz、15分の低頻度連続刺激はin vivoのラット大脳では長期抑圧を起こさないことを見出した。 2)in vivoの脳に存在する内因性の脳由来神経栄養因子が上述したシナプス長期抑圧誘発の阻止に関与していることを発見した。 3)脳発達における新規Postsynaptic density(PSD)足場蛋白質PSD-Zip70の発現を検討したところ、神経前駆細胞からニューロン分化への最も早い時期に発現すること、及び神経前駆細胞の細胞増殖を抑制することを見出した。 4)網膜の生後発達におけるリソソームカテプシンDの役割を検討した結果、カテプシンDを欠損するとphotoreceptor細胞がアポトーシスで死滅し網膜色素症を呈すること、一方内顆粒層の細胞はミクログリアに誘導される一酸化窒素(NO)合成酵素を介してネクローシスで死ぬことが判明した。
