老化促進マウス(Klothoマウス)の脳機能解析

研究概要	遺伝子改変老化促進マウス(Klothoマウス)は,多彩なヒト型の老化徴候を示し,生後約100日で老化により死亡する.原因遺伝子であるKlothoは脳と腎臓で高発現しており,脳機能に深く関与していると推定される.本研究では,Klothoマウスの運動機能および記憶・学習能力などの高次脳機能の変化とそのメカニズムについて行動学的および神経化学的に解析した. 運動機能および運動学習能の評価としてspontaneous locomotor activity試験,rota-rod試験を行った.6週齢のKlothoマウスの自発運動量は野生型と比較し変化していなかったが,7週齢では有意に減少していた.6から8週齢のKlothoマウスでは,rota-rodから落下するまでの時間が野生型に比べ短縮していた.学習・記憶能力の評価としてconditioned-fear試験およびnovel-object recognition試験を行った.Klothoマウスではconditioned-fear試験において連合学習能力に,novel-objectrecognition試験において認知記憶能力に障害が認められた.酸化的ストレスの消去系に関与している脳内のCu/Znスーパーオキサイドディスムターゼ(SOD)活性およびグルタチオン(GSH)含量を測定した.KlothoマウスのCu/Zn SOD活性およびGSH含量は野生型マウスに比べ有意に減少していた. Klothoマウスでは野生型に比べ運動機能,連合学習および認知記憶能力が低下し,脳内Cu/Zn SOD活性およびGSH含量が低下していたことから,Klothoマウスに認められた行動変化には酸化的ストレスが関与している可能性が示唆された.
