研究概要	1.大脳皮質出力ニューロンを逆行性に細胞体・樹状突起をGolgi染色様に標識する方法により、前もって出力ニューロンを標識したラットの大脳皮質スライス標本を作成した。微小ガラス電極を各層のニューロンに刺入し、基本的な電気的性質を記録してニューロンを分類し、Biocytinを注入した。スライス標本を固定した後、組織学的な検索として免疫蛍光法を用いてBiocytinを注入したニューロンが興奮性あるいは抑制性であるか検討した。次に、Biocytinを注入したニューロンと特にその出力である神経軸索をAvidin-biotinylated peroxi-dase complex法によって青黒色に染色し、逆行性に標識された出力ニューロンの樹状突起をPeroxidase anti-peroxi-dase酵素免疫染色法で赤く染色し、1個のニューロンから一群の大脳皮質投射ニューロンへの連絡を調べた。その結果、興奮性のIII層錐体ニューロンの出力はV層の皮質脊髄投射錐体ニューロンへ多く入力しているが、VI層の皮質視床投射錐体ニューロンへはその4分の1しか入力していないことを見出し報告した。さらに、VI層の錐体ニューロンへの入力を調べII/III層及びV層の錐体ニューロンからの入力は少ないが、IV層・VI層からの入力が多いことを見いだしており、報告をまとめている。 2.中枢神経系の投射ニューロンをアデノウィルスベクターあるいはSindbis virusベクターに感染させ、膜移行シグナルを導入したGreen fluorescent protein(pGFP)を発現させ、Golgi染色様に標識することに成功した。 3.GABA作動性のインターニューロンをGolgi染色様に染色する手法を、トランスジェニックマウスを利用して作ろうとしている。現在、GABAニューロンのマーカーであるParvalbuminとCalretininというカルシウム結合タンパク質の遺伝子をBACを用いてクローニングしている。Calretininトランスジェニックマウスは作成済みで、pGFPの発現特異性を検討している。
