視交叉上核の発育と視神経投射、光同調機構についての先端的研究

研究概要	哺乳類の生体リズムの中枢である視交叉上核(SCN)への視床下部外からの神経入力、とくに網膜神経節細胞からの視神経投射と光情報によるリズム同調機構に注目して研究を進めている。SCNへの視神経入力は生後2-3wにかけて急増しピークに達する。ラットでは視神経はSCNの腹外側部に投射し、我々はこれまで腹外側部に存在するVIPニューロンやGRPニューロンに直接シナプス入力することを報告してきた。SCN腹外側部に一致してアストログリアも存在し、出生時に両眼を摘出するとSCNでのアストログリアのマーカーであるGFAPの染色性がコントロールに比べると2-3wにかけての増加が見られず、SCNへの視神経入力がGFAP陽性のアストログリアの突起伸長に必要なことが分かった。次に出生時に両眼を摘出すると他の神経核外からの入力、外側膝状体からのNPY線維の投射はが増加することが分かり、一方外側膝状体のNPYニューロンの細胞数を数えると増加がみられず、NPY陽性軸索の分枝が増加したためと考えられた。そこで視神経入力が2-3wにかけて急増する原因についての検索をはじめている。まず既知の神経ガイダンス因子のSCNでの発現探索について、PCRによる検索を行った。生後2W、adultで軸索の反撥因子であるslit1,2,ephrin-A5の発現がPCRで検出されたが、両者で発現量の差は見られなかった。次に2WのとadultのSCNについて遺伝子発現に差があるものをPCRサブトラクション法で検索をはじめ、2WSCNで特異的に増加している候補遺伝子が数種類上がってきていて、現在ノーザンブロットやPCR、in situ hybridizationで発現差を確認中である。
