神経伝達物質の合成と老化・神経細胞死との関わり

研究概要	本研究は、カテコールアミン生合成律速酵素であるチロシン水酸化酵素遺伝子を、誘導的遺伝子破壊法を用いて成獣マウスの脳で任意の時点に破壊することにより神経伝達物質を合成できなくなったニューロンが、神経細胞死を起こすかどうかを調べることを目的としている。この研究は、老化によって起こる神経伝達物質の減少がアルツハイマー病やパーキンソン病における選択的細胞死に関わっているかを知ることができるため、これらの疾患と老化との関連を探るために重要な知見を与えるものと期待される。DNA組換え酵素Creに、エストロゲン受容体のリガンド結合部位を融合させたCre-ERTを用いることにより、DNA組換え活性を薬剤で誘導可能とした。これまでに、チロシン水酸化酵素遺伝子のイントロン内にloxP配列を導入したマウスの作製と、Cre-ERTをチロシン水酸化酵素のプロモータ制御下に発現するマウスの作製に成功した。Cre-ERTの発現マウスは、チロシン水酸化酵素の第1エキソンにin-frameでCrc-ERTを挿入したCre-ERTノックインマウスと、チロシン水酸化酵素遺伝子のプロモータにCre-ERTをつないだトランスジェニックマウスの2種を作製した。今後Cre-ERT発現マウスでのCreの発現分布について解析するとともに、loxPマウスとかけ合わせて、薬剤誘導的にチロシン水酸化酵素を破壊して神経機能に対する影響を調べる。チロシン水酸化酵素を破壊された神経細胞が、伝達物質を合成できない状態で生存し続けることができるかどうかについて検討する。
