研究概要	胃がんの約一割の症例ではすべてのがん細胞にEBウイルス(EBV)が感染している。本研究は、胃がんにおけるEBVの役割解明を目的として研究を行い、以下のことを明らかにした。 1.EBERは上皮細胞由来Intestine407細胞においてFAS誘導性のアポトーシスへの抵抗性を誘導した。さらに、EBERがインターフェロンによって誘導される蛋白キナーゼPKRに結合し、PKRの活性化阻害を起こすことを明らかにした。ドミナントネガティブPKRを用いた解析の結果、EBERはPKRと結合しそのキナーゼ活性を阻害することによりFasを介するアポトーシスに対する抵抗性に寄与することが示された。 2.EBVがコードするBARF1はげっ歯類の線維芽細胞を悪性変換するなど、がん遺伝子としての活性が知られている。BARF1は従来EBV産生に伴って発現する溶解感染遺伝子群の1つで、大部分のがん細胞では潜伏感染状態にあるためBARF1は発現していないと考えられていた。本研究ではリアルタイムPCR法による解析の結果、BARF1がEBV関連胃がん、上咽頭がんにおいて高いレベルで発現していること、さらに、他の溶解感染遺伝子がほとんどあるいは全く発現していない状態でBARF1が高度に発現していることを明らかにした。以上の結果、BARF1がEBV関連胃がんの発生に深く寄与していることが示唆された。
