腫瘍拒絶におけるヘルパーT細胞の認識抗原および機能制御分子の解明

研究概要	癌抗原ペプチドの発見は、腫瘍免疫学の発展に大きなインパクトを与え、癌特異的免疫療法の実現性に道を開いた。しかし、欧米のクラスI結合性癌ペプチド単独投与の臨床研究では、未だ期待されたほどの効果は認められていない。我々は、担癌生体における免疫抑制を打破して癌特異的CTLを誘導するために、いくつかのストラテジーを開発してきた。本年度は、ヒト樹状細胞、Th1細胞治療の抗腫瘍メカニズムと遺伝子改変癌特異的Th1細胞の加工法の開発を中心に研究を行い以下の結果を得た。 (1)ヒト末梢血単核球をGM-CSF+IL-3+Th1サイトカインで刺激することにより抗腫瘍免疫誘導に好適な樹状 (2)抗原特異的Th1細胞の移入と癌抗原タンパクの腫瘍内投与によって、MHC class II^-の腫瘍も完全拒絶することができる癌治療モデルを確立した。 (3)Th1細胞療法をより普遍的な癌治療にするために、遺伝子改変Th1,Tc1の細胞加工法も確立した。すなわち、癌特異的マウスTh1のTCR遺伝子を導入することによって非特異的なTh1に癌特異的性を付与することに成功し、in vivoでも強い抗腫瘍効果を示した。 (4)抗体とTCRシグナルキメラ遺伝子を導入することによってMHC非拘束的に作用できる遺伝子改変癌抗原特異的Th1細胞の加工にも成功した。すなわち、CEA特異的単鎖抗体遺伝子をCD28,CD3ζ遺伝子のシグナル伝達部位に結合させたcIgTCR遺伝子をレンチウィルスベクターを用いて非特異的に活性化させたTh1,Tc1細胞に導入することにより、癌特異性を付与することに成功した。以上の結果より、Th1細胞治療は広く応用可能な癌免疫療法になると考えられる。
