S6キナーゼを中心とする翻訳制御の細胞増殖,発がんにおける関与

研究概要

1.新規のS6キナーゼ(S6キナーゼ2)欠損マウスの作製
S6キナーゼ2ゲノムがターゲットされたES細胞を用いてキメラマウスを作製し、本遺伝子欠損マウスの作製に成功した。個体レベルで欠損マウスと野生型マウスの間でそのフェノタイプに大きな差はなかった。またTリンパ球、胎児由来繊維芽細胞の増殖速度は野生型とものと明らかな差は認められなかった。
2.S6キナーゼを制御するホスファターゼの解析
S6K1に結合し、直接その活性を制御している2A型ホスファターゼ(PP2A)の阻害蛋白cDNAのクローニングを行った。新規のものを含めて3種類の分子種を同定した。これら3種類のものは全く異なった組織分布をしていたが、いずれもG1/Sでその発現が上昇し、肝がん細胞で過剰発現をしていた。
3.翻訳制御機構の解析
mRNAのプロセッシングに働くNIPP-1蛋白質の様々な変異体を用いた実験により、この蛋白がAキナーゼや1型ホスファターゼ(PP1)を介して翻訳の調節に働いていることが示唆された。
4.S6キナーゼシグナルとMAPKシグナルの関係
MAPKのうちのJNKの脱リン酸化/不活性化に特異的に働くホスファターゼの単離に成功し、その解析をおこなった。このホスファターゼはホスホチロシン、ホスホセリン/スレオニンの両方を脱リン酸化する2重特異性ホスファターゼに属し、核-細胞質輸送に関係するシャトル蛋白であった。
