皮膚癌を誘発する紫外線からの新規抗酸化酵素ペルオキシレドキシンによる防御機構

研究概要

紫外線は皮膚癌を誘発する大きな要因であり、それには活性酸素が関わっている。本研究ではヒト皮膚ケラチノサイト由来のHaCaT細胞とマウスメラノーマ由来のB16-F1細胞を用いて、紫外線による新規抗酸化酵素ペルオキシレドキシン(Prx)の発現誘導作用とp53の活性化に対する作用について検討した。紫外線を照射すると、B16-F1細胞において照射量依存性にサイクリン依存性キナーゼ阻害タンパク質であるp21が発現誘導された。これはp53が変異型であるHaCaT細胞では見られなかったことからp53に依存した発現誘導であると考えられた。p21の発現誘導は60mJ/cm^2でピークを示し、その後低下した。p21の発現誘導は放射線照射によっても観察されたが、この場合には1200Rという高照射量でも発現誘導の低下は見られなかった。同様の系で紫外線と放射線を照射した際のペルオキシレドキシン(Prx)の発現誘導を検討した。Prxには、I〜VIのアイソフォームがあるが、検討の結果メラノーマ細胞ではほとんどのアイソフォームが発現していた。今回は細胞に最も豊富に存在するPrxIと、KGFで発現が誘導されるPrxVIについて詳細に検討した。その結果、PrxIはどちらの細胞においても発現が誘導されなかった。また細胞内では細胞質に局在するが、紫外線照射により核に移行するという現象も観察されなかった。一方、PrxVIはB16-F1細胞を放射線照射した時に発現誘導が見られた。この際照射量依存的にp21の発現誘導も認められた。また代表的な抗酸化酵素であるCu/Zn-SODはHaCaT細胞における紫外線照射で誘導が見られなかった。これらの結果から、皮膚の細胞が紫外線や放射線障害に対して細胞を守る機構のひとつとして、p21とPrxを発現誘導させて細胞周期を停止させ、かつ活性酸素を消去する機構が存在することが示唆された。
