HTLV-I Taxによる細胞周期転写因子E2F活性化の分子機構

研究概要	成人T細胞白血病(ATL)はHTLV-Iの感染に起因する。HTLV-Iの産生する蛋白のうちTaxがT細胞白血病化に主要な役割を果たしている。Taxは、T細胞の増殖情報伝達に関わる因子や遺伝子に作用することが知られている。しかし、分子機構にはいまだ不明な点が多く、現在の知見はT細胞の腫瘍化を系統的に説明できるものではない。我々はTaxが細胞周期転写因子E2Fを活性化すること、ヒトT細胞の細胞周期をS期まで進めることを見い出している。 IL-2依存性ヒトT細胞株Kit225細胞に組換えアデノウイルスを用いてTaxを発現させ、TaxによるE2F活性化分子機構を解析して次の結果を得た。TaxはE2F1の転写を促進しGl/S期でのE2F1の量が増大させる。また、Taxの発現により活性型の遊離型E2F4が誘導される。これはTaxによるCdk2、Cdk4のリン酸化酵素の活性化に起因している。Cdkの活性化は、Cdk遺伝子と、Cdkに結合するサイクリンD2、サイクリンEの遺伝子の転写レベルでのTaxによる促進が主因と考えられる。E2F1、Cdk、サイクリンの各遺伝子のTaxによる発現誘導は、細胞周期が進行する前に認められ、Taxによる直接の作用である。これらの結果は、Taxによる転写促進作用が細胞増殖促進に直接結び付いていることを示していて、HTLV-Iによる白血病発症の分子機構の解明に大きく貢献できたと考えられる。
