テロメア機能異常を介した染色体異常の発生機構

研究概要

テロメレースをもたない正常細胞は、細胞分裂のたびに末端複製問題によってテロメア末端が短小化し、テロメア長がある閾値に達すると細胞老化表現型が誘導される。細胞老化は、非可逆的な細胞増殖の停止とともに細胞質や核の巨大化などの特徴を示す状態であるが、上記のテロメア短小化による複製老化のほかに、種々のストレスによっても誘導されることが知られている。そのような細胞老化誘導刺激の中には、酸化ストレス、不適切な細胞培養環境、および活性化Rasがん遺伝子の過剰発現による過剰な増殖刺激などが含まれる。しかし、これらのさまざまな刺激による細胞老化誘導系において、共通の分子機構が関与するか否かは不明であった。今年度の本研究において、我々は、ストレス誘導性MAPK(Mitogen-activated protein kinase)p38の活性化が重要な役割を果たしていることを明らかにした。特に、p38の上位キナーゼMKK6の過剰発現によって、人為的に細胞老化を誘導できることを示したほか、p38を薬理学的あるいは遺伝学的に機能抑制することで、細胞老化の誘導を少なくとも部分的に回避することができることを示した。さらに、p38の活性化が細胞増殖停止を誘導するにあたり、Rb蛋白質が必須な下流分子の一つとして機能するが、p53は必須ではないことを示した。このように様々な刺激による細胞老化誘導が保存された経路で行われることは、細胞老化が適応的な役割をもつことを示唆している。特に、細胞老化のがん抑制機構としての役割が注目された。
