腫瘍退縮に至る担がん宿主T細胞の腫瘍局所浸潤の分子機構

研究概要	担癌宿主リンパ組識内で腫瘍特異的T細胞応答が検出されても、必ずしも腫瘍が免疫退縮を遂げるとは限らない。腫瘍拒絶に至るには抗腫瘍T細胞の腫瘍局所への浸潤が不可欠である。IL-12投与により治療効果が認められる腫瘍系では、腫瘍局所に於いてT細胞上の接着分子LFA-1/VLA-4と腫瘍周辺血管内皮細胞のICAM-1/VCAM-1との相互作用を介して著明なTh1型T細胞の浸潤が誘導される。本研究では、IL-12によるT細胞浸潤の増強機構を解析した。T細胞抗原受容体での刺激により活性化されIL-12受容体を発現したT細胞は、IL-12刺激に応答してケモカイン受容体CCR5の発現を誘導した。このT細胞が腫瘍塊内に存在するMIP-1α/βで更に活性化されるとLFA-1分子のICAM-1分子への接着能が著明に増強した。また、腫瘍内T細胞浸潤はケモカイン受容体CXCR3よっても支配されていた。即ち、CXCR3はIL-12刺激の結果産生されるIFN-γによりT細胞自身に誘導され、これにIP-10が作用することでLFA-1分子のICAM-1分子への接着能が増強された。なお、CCR5発現は転写因子STAT4依存性に、一方CXCR3はSTAT4非依存性に誘導された。一方、CCR5特異的阻害剤として開発されたTAK-779は、Th1細胞に発現されるCCR5とCXCR3両受容体に対して選択的な阻害効果を示すことを明らかにした。興味深いことに、TAK-779をin vivo投与するとIL-12治療によるT細胞浸潤が抑制されるとともに、腫瘍退縮効果もほぼ完全に阻害された。以上、抗腫瘍性T細胞の腫瘍局所浸潤機構の分子機構の解明により、担癌宿主抗腫瘍T細胞の腫瘍塊内での拒絶反応に至る詳細な過程が明らかとなった。これらの知見は腫瘍免疫療法の効果判定に関する分子レベルでの予知指標を提供する。
