ヒトCD4^+T細胞が認識する腫瘍特異抗原の同定

研究概要	膵癌を対象としてCD8陽性T細胞およびCD4陽性T細胞が認識する腫瘍拒絶抗原エピトープを同定することを最終目的として、既に昨年度の研究により癌患者血清中の抗腫瘍抗体を用いたアロ膵癌細胞株由来cDNA発現ライブラリーのスクリーニング(SEREX法)により、膵癌抗原遺伝子として既知の遺伝子13種類、および未知の遺伝子5種類を同定した。過去にSEREX法を用いて多くの癌抗原が同定されているが、そのほとんどについて細胞傷害性T細胞(CTL)に対する抗原性は確認されていない。そこで本年度は、上記の膵癌抗原がCTLに認識されるか否かを調べた。解析した6種類の癌抗原のうちCLPおよびKM-PA-2の2種類が、大腸癌組織に浸潤するリンパ球(TIL)由来のCTL株にHLA-A2拘束性に認識され、それぞれについてCTLが認識する抗原ペプチドを3種類づつ同定した。これらのペプチドを用いて癌患者および健常人の末梢血単核細胞(PBMC)より、HLA-A2拘束性かつ癌細胞を特異的に認識して破壊するCTLを誘導できた。またCLPあるいはKM-PA-2のCTLエピトープに反応するIgGおよびIgE抗体が、癌患者のみならず健常人の血清中にも検出され、抗体陽性者ではin vitroにおける腫瘍抗原反応性CTLが樹立されやすい傾向が認められた。さらに、これらのCTLエピトープと相同性が高い微生物由来の非自己抗原ペプチドが存在し、双方のペプチドの間にCTLおよびIgG/E抗体の交叉反応性が観察された。これらの結果は、1)腫瘍免疫において同一の抗原ペプチドが細胞性ならびに液性免疫系に共に認識されうること、ならびに2)感染免疫と腫瘍免疫の間に、交叉反応性が存在する可能性があることを示すものである。
