ヒトT細胞が認識する腫瘍特異抗原の同定

研究概要	本研究はSEREX法あるいはcDNAマイクロアレイ解析データを用いて、腫瘍免疫において最も有効な標的と考えられる癌特異的に高発現している抗原を選び出し、これらの抗原についてヒト細胞傷害性T細胞(CTL)およびヘルパーT(Th)細胞に、腫瘍拒絶を誘導するペプチドを同定することを目的とする。本年度の成果として、当初の目的にかなう3種類の腫瘍特異抗原、KM-HN-1、GPC3およびHSP105に加え、新たにPP-RPを同定した。これらの遺伝子の情報をもとに抗原ペプチドを合成して、癌患者の末梢血単核細胞(PBMC)より、これを特異的に認識するヒトT細胞株を樹立し、これが当該抗原を発現する腫瘍細胞株を傷害することを明らかにした。HSP105については蛋白抗原を用いて、大腸癌患者のPBMCより複数のHSP105特異的Th細胞株を誘導できた。またKM-HN-1に対する血清中のIgG抗体が健常人には認められないが、胃癌、肺癌、大腸癌など様々な癌患者において高頻度に観察されることを見いだし、癌細胞傷害性CTLを誘導できるKM-HN-1ペプチドを同定した。さらにGPC3に関しては、マウスのin vivoにおいても腫瘍拒絶抗原として機能し、抗腫瘍免疫応答を誘導することを確認した。また、遺伝子導入により抗原とケモカインを共に発現させたマウスES細胞由来の樹状細胞が、当該抗原に特異的な強い抗腫瘍免疫応答を誘導し、特にSLCは強い効果を示すことを明らかにした。
