転移性上皮癌ワクチン開発の基礎的研究

研究概要

食道癌患者末梢血より樹立した細胞傷害性T細胞(KE4-CTL)を用いて新たにクローニングした遺伝子の塩基配列がlckと同じであった。本遺伝子産物のp56^<lck>は、リンパ球系細胞の増殖や機能発現に関与する事はよく知られているが、転移性大腸癌や肺癌の一部でも異所性に発現している事から、本分子が転移癌と関連した癌ワクチンの候補になり得るかどうかを検討した。(a)lckは正常細胞・組織ではType IIプロモーターが関与しているのに対して、癌細胞では主にType Iが関与していることがRT-PCRで確認され、タンパク質レベルでは両者共同じ易動度のタンパク質としてウエスタンブロット法で検出された。(b)タンパク質の発現は正常細胞でも認められたがKE4-CTLは認識しなかった。しかし、癌細胞と正常細胞に対するCTLの認識の違いに関する機序に関しては現在のところ明らかではない。(c)HLA-A24結合モチーフを有するペプチドを合成し、C1R/A*2402細胞株と反応させた後CTLが産生するIFN-γを測定し、KE4-CTLがHLA-A24拘束性に認識するペプチド抗原を3種類(Lck208-218,Lck486-494およびLck488-497)同定した。次に、患者末梢血PBMCをこれらの3種類のペプチド抗原で刺激し、限界希釈法でCTLの誘導頻度を計算で求め、CTL前駆細胞の割合を予測した。KE4細胞株に特異的なCTLのin vitroでの誘導を試みたところ、転移癌患者ではペプチド刺激していない群でも前駆細胞が認められ、刺激することにより増強されることが示唆されたが、非転移癌患者および健常人では前駆細胞出現頻度の増強は認められなかった。以上の実験成績から、本ペプチドが、特に転移癌の癌ワクチン候補となりうることが示唆された。
