研究概要	(1)4種類の変異型Tsc2遺伝子を導入したTransgenic Eker ratを作成し、変異型Tsc2遺伝子導入Transgenic Eker ratのmutantホモ接合体の致死の回避の有無、ヘテロ接合体の腎発がんの抑制の有無を調べた。興味あることに、胎性致死と腎発がんにおける機能ドメインの解離が見られた。 (2)Tsc2遺伝子knockoutマウスとTsc1遺伝子knockoutマウスの解析と交配実験を行った。Tsc1遺伝子knockoutマウスの表現型は、Tsc2遺伝子knockoutマウスと、良く似ているが興味あることに、Tsc1遺伝子knockoutマウスの腎発がんの方が軽度であった。共に、肝臓に血管腫が高率に発生した。現在、Tsc1遺伝子knockout miceとTsc2遺伝子knockoutマウスを交配させ、肝腫瘍で組織発生の観点から、興味ある知見が得られた。 (3)Tsc2癌抑制遺伝子の発現制御機構の解明を行った。腎がんで発現が亢進し、且つcEts結合配列に結合する細胞蛋白の単離、同定をsupper gel shift assayおよびEker腎癌細胞のcDNAライブラリーを作製しSouthwestern法にて試みた。Etc family蛋白のTsc2癌抑制遺伝子の発現制御における重要性が確認された。 (4)多段階発がん過程でのErc遺伝子と新規Niban遺伝子の発現解析を行った。Erc遺伝子産物、Niban遺伝子産物のペプチドおよびモノクローナル抗体の作製と発現を調べた。共に、ヒト腎癌でも発現がみられ、Erc遺伝子産物は、分泌型でもあり、診断的腫瘍マーカーの可能性もある。Niban遺伝子産物は、極く初期の病変から発現が見られた。
