マウスモデルを用いた造血器腫瘍原因遺伝子の研究

研究概要	ヒト骨髄性白血病原因キメラ遺伝子NUP98-HOXA9トランスジェニックマウスにレトロウィルス挿入変異実験を行い白血病発症に重要な5種類の協調遺伝子候補を同定した。NIH 3T3細胞を用いた実験により、これらの遺伝子がNUP98-HOXA9の形質転換能を促進することが明かとなった。骨髄性白血病及びB細胞性白血病原因遺伝子Evi9/BCL11Aの遺伝子破壊マウスを用いて、Evi9がpre-B細胞分化に必須であることを明らかにした。また、Evi9 AZ isoformを強発現させた骨髄細胞の移植がEvi9 null変異を持つ胎児肝細胞同様T細胞性白血病の発生を促すこと、この白血病が宿主由来であることを明らかにした。さらに、Evi9のin vitroにおけるE1及びE2 ligaseの存在下でのSUMO化を確認し、SUMO1とEvi9とを発現させるとSUMO1の核内における局在に再構成が生じることを確認した。新規リンパ腫抑制遺伝子・Rit1の遺伝子破壊マウスを用いて、γ線照射による発がん実験と生じたリンパ腫での遺伝子変異を検討した。Rit1(KO/+)マウスはRit1(+/+)マウスに比べ、明らかに高い頻度で胸腺リンパ腫を発症することが示された。Rit1(KO/+)マウスに発症したリンパ腫の約50%に野生型のRit1アレルのLOHを認めた。一方、V(D)J組換えによるRit1内部欠失は検出されなかった。これらの結果からRit1遺伝子が、がん抑制遺伝子であることが確認された。Rit1蛋白の機能解析を試み、細胞増殖とアポトーシスとの関連性が示唆された。胸腺T細胞で特異的に発現するLckプロモーターを用い、Rit1βWT(野生型)とRit1βKT(核移行に障害をもつ変異型)のトランスジェニックマウスを作製し、異所性Rit1遺伝子発現が個体の発生に障害をもたらす可能性が示唆された。
