環境中の発がん物質により誘発される遺伝子変異の特異性と遺伝子発現解析

研究概要

加熱魚肉食品中に含まれるHCA類の一つPhIPにより誘発されるラット大腸がんモデルを用いて、発がん感受性の異なるF344系統とACI系統における大腸上皮での遺伝子発現プロファイルについて、GeneChip(Affymetrix社)を用いた包括的発現解析を行った。PhIP投与直後において両系統間で発現誘導される遺伝子の違いが認められた。PhIP投与終了後42週においては、がん部での系統差は軽微であったのに対し、非がん部では200個以上の遺伝子に2倍以上の発現差を認めた。高感受性のF344系統では、リボゾームRNA、リボゾーム蛋白などの転写・翻訳に関わる遺伝子の発現が高かった。脂肪の輸送に関わるL-EABP、I-EABP、α-Synuclein遺伝子などの発現も有意に高く、高カロリー・高脂肪食と大腸発がんとの関連性において興味ある結果である。低感受性のACI系統では、ミスマッチ修復遺伝子Msh2や、Comt, Gst mu1などの一部の解毒・代謝関連遺伝子の有意な発現上昇が認められた。これら遺伝子の発現量や誘導性の違いが、PhIPに対する発がん感受性を規定している可能性がある。さらに、種々のHCAおよびAOMによる大腸前がん病変ACFの誘発性と遺伝子発現プロファイルについても検討した。従来より、AOM, PhIP, IQ, Glu-P-1の大腸発がん性はほぼ同等であり、MeIQはやや低いと考えられているが、今回の実験結果では、ラット一匹当たりのACF誘発数はそれぞれ135±22個、2.7±1.5個,1.8±0.8個,1.8±1.3個,5.6±2.3個であった。さらに、大腸発がん性のないとされるMeIQxでも平均1.6±1.1個と、IQ, Glu-P-1とほぼ同等のACF誘発数を示し、発がん性とACF誘発性とが必ずしも相関しない事が分かった。また、AOMとPhIPはほぼ同等の発がん性を示すものの、ACF誘発数や発現遺伝子のプロファイルには大きな差異があることから、両者の発がんプロセスが大きく異なることが示唆された。種々のがん原性物質により惹起される遺伝子の発現変化を解析することにより、各化合物による発がん分子機構の相違点や大腸発がんに重要な役割を果たす遺伝子を同定できる可能性がある。これら包括的な遺伝情報を集積し、発がん性との関連性を詳細に検討することにより、既知および未知の化合物の大腸発がん性を早期段階で予測できる可能性が期待される。
