がん細胞の酸素・栄養飢餓抵抗性の機構とその進展における役割

研究概要	栄養飢餓耐性の膵癌細胞PANC-1にAMPKα1,α2のアンチセンスRNA発現ヴェクターを安定導入すると、栄養飢餓耐性が失われた。これらの細胞は同時に、軟寒天培地でのコロニー形成能、ヌードマウスでの腫瘍形成能も失っていた。一方、HepG2や正常線維芽細胞を用い、AICARにてAMPKを活性化するとグルコース欠乏耐性となった。このときAMPK a2 subunitのアンチセンスRNA発現ヴェクターを安定導入するとこの反応が起こらないことから、AMPK a2の活性化によることが解った。また、HepG2や正常線維芽細胞をグルコース飢餓にすると細胞回転はG1で停止する。この反応は基本的にはp53依存性で、p53のセリン15番の強い燐酸化が起こりp21waf1の増加を引き起こす。P53の燐酸化はAMPKの活性化剤で起こることからAMPKが関与している可能性がある。さらに、同様の反応はataxia teleangiectasis患者の線維芽細胞では起こらないこと、ATM発現ヴェクターで回復すること、HepG2細胞ではATMのアンチセンスRNA発現ヴェクターで抑制されることからATM依存性であることが解った。
