研究概要	わが国の癌死亡原因の第1位である肺癌を、一つの『疾患』として統合的な理解することを目指し、多面的な分子病態解析研究を展開し下記の成果を得た。 (1)ヒト肺癌の遺伝子発現プロファイル解析を進め、これまで比較的均質な腫瘍と考えられてきた肺扁平上皮癌が、臨床病理学的に際立った特徴を持つ2群にクラスタリングされ得ることを示すなど、癌の個性解明につながる成果を得た。また、外科切除から5年後の生存の可否について、とくに非扁平上皮癌においては90%以上の高い確度で予測することができるシステムの開発にも成功した。 (2)我々が樹立した高転移性ヒト肺癌細胞亜株(NCI-H460-LNM35)を用いた網羅的遺伝子発現解析を通じて、高転移能獲得に伴って親株に比して有意な発現上昇を示すとともに、高転移性ヒト肺癌細胞亜株の高い運動能や浸潤能の付与への機能的な関与を示す新規遺伝子LNM01遺伝子を同定することに成功した。LNM01遺伝子の高発現は、ヒト肺癌症例におけるリンパ節転移の有無や外科切除後の生命予後などの臨床病態に有意な関連を示した。 (3)肺癌の分子病因としての細胞周期制御機構の破綻に関する検討は、ヒト肺癌細胞株においてdecatenation G2チェックポイント異常がみられることを初めて明らかとし、高度な染色体不安定性の獲得要因となっている可能性を示唆する結果を得た。
