がん患者の抗体を用いた発現クローニング法(SEREX)によるがん抗原の同定

研究概要	癌患者の免疫系が抗原として認識する自己の癌細胞に発現する分子を同定すること、またそれらの中から癌の免疫診断・治療の標的となりうる癌抗原を選択することが、本研究の目的である。その方法として、担癌患者が産生するIgG自家抗体をプローブとして自己癌組織由来の発現cDNAライブラリーを検索し癌抗原を同定するSEREX法を用いた。本法の利点は癌細胞の培養株の樹立を必要としないことであり、あらゆる癌で癌抗原を検索することが可能である。これまで胃癌5症例、前立腺癌3症例、乳癌2症例、大腸癌2症例、悪性軟部腫瘍2症例、肺癌1症例の検索を行った。癌患者の免疫系は予想外に活発であり、分泌蛋白、細胞膜蛋白、細胞質内蛋白、核内蛋白など700種を越える抗原を同定した。これらの中から診断・治療に有用な抗原を選択するために、各々の抗原の発現特異性と抗原に対する免疫応答の癌特異性を検討した。発現特異性に関しては、まずUniGene Cluster Databaseでの検索で推定し、有望と考えられた抗原に関してはさらにRT-PCRとノーザンブロットで解析した。広範な組織に発現する抗原がほとんどであったが、癌が発生した組織を含め極めて限局した組織で発現する抗原ならびに癌で発現増大する抗原も同定された。さらに、抗原に対する免疫応答の癌特異性、すなわち抗体産生の癌患者特異性を検討した。複数の癌患者で抗体が検出されるが非癌患者コントロール群ではされない抗原と、癌患者群における抗体検出頻度が非癌患者群と比較して圧倒的に高い抗原が有望な抗原である。これらの検討で、CT(Cancer Testis)抗原、分化抗原、癌で発現増大する抗原、内在性レトロウイルス等の約60種の候補抗原が選択された。現在、選択した抗原を免疫診断・治療に利用するために、抗体検出の高感度化、細胞傷害性T細胞誘導能等を検討している。
