造血微小環境による造血幹細胞の分化制御機構

研究概要	多能性造血幹細胞の間質細胞による分化列決定の制御機構を明らかにするため、間質細胞依存に未分化状態を維持したまま増殖する造血幹細胞様細胞株THS119細胞をモデル系として用い、未分化造血細胞の間質細胞依存性増殖の制御機構を解析した。まず、未分化な血液細胞の間質細胞依存性制御からサイトカイン依存性の制御に転換してゆく過程の細胞内シグナル伝達機構について、間質細胞依存性に増殖する未分化血液細胞株THS119細胞と、この細胞から間質細胞非依存性、サイトカイン(IL-7、IL-3)依存性に増殖可能になった細胞株(THS119/IL-3,THS119/IL-7)を用いて、間質細胞ならびにサイトカイン依存性の増殖におけるシグナル伝達経路の相違について検討し、両者で、STAT5経路の活性化(STAT5のリン酸化)が異なること、間質細胞依存性増殖では、anti-apoptotic遺伝子であるbcl-2の誘導が認められるが、サイトカインはこの遺伝子を誘導できないこと、STATファミリーの中、間質細胞依存の増殖においては、STAT1蛋白質の誘導が特異的に認められること、などを明らかにした。さらに、間質細胞株依存的に増殖する造血幹細胞株DFC-28の間質細胞依存的な分化誘導の解析から、間質細胞株の選択的支持機能に依存して、その分化の過程が可逆的に制御されることを実験的に始めて明確に示した。これらの結果から、骨髄のような複雑な造血環境の中では、接着する間質細胞によって未分化血球の分化段階か可逆的にスイッチされるものと予想され、今後、この闘質細胞が示す分化の可逆的誘導がどのような機能分子によるのかを明らかにしたい。
