研究概要

がん細胞における細胞周期制御異常の機構を明らかにすることを目的として、大腸癌のがん抑制遺伝子APC、Wntシグナルの機能を解析し以下のような知見を得た。
1.APCとAsefは細胞密度や増殖因子刺激に応じて細胞接着面、細胞質、細胞運動の先端部分(先導端)、ラッフリング膜、核など様々に局在を変化させ、細胞接着、細胞運動に関与することを明らかにした。
2.β-catenin-cadherin複合体に結合するGAP分子RICSは、Cdc42およびRacを標的とし、Srcファミリーのチロシンキナーゼによりリン酸化を受けることを見出した。
3.癌遺伝子BCL9の産物と相同性のあるB9Lが、Pygoおよび未知の因子と結合してβ-catenin-TCF複合体による転写活性化を促進し、細胞transformationを促進する作用をもつことを明らかにした。B9Lが大腸癌の30%の症例で異常発現していることを見出した。
4.TGF-βの擬受容体BAMBIがWntシグナルの標的で、TGF-βによる増殖抑制を解除する作用があること、大腸癌、肝癌の70%程度で過剰発現していることを見出した。
5.Wntシグナルの負の制御因子DKK1が、β-catenin-TCFの直接の標的遺伝子で、負のフィードバックループを形成していることを見出した。
6.DLGがRap1特異的なGAP分子SPALと結合してシグナル伝達の足場として機能することを明らかにした。DLGホモ欠損マウスは誕生直後に死に、ヘテロ欠損マウスは癌を発症した。SPALホモ欠損マウスの一部は生後1ヶ月ほどで死亡した
