PI3K-Akt経路による癌化メカニズムの解析

研究概要	前年度までに癌原遺伝子産物Aktが繊維芽細胞の増殖因子依存的な細胞運動の促進に主要な役割を果たすこと、またAktが低分子量Gタンパク質Racの下流で活性化し、Racによる細胞運動能の上昇に関与することを報告した。しかしながらRacがいかなるメカニズムでAktを活性化しているかは明らかではなかった。本年度において、Racによって活性化されることの知られていたPAKがAktの活性化に重要な役割を果たしていることを見出し、その活性化メカニズムの一部を明らかにした。最近子宮癌種においてPAK1を含む染色体のある領域が増幅されており、さらにその中でPAK1が原癌遺伝子として必須の役割を果たしているという報告がなされているので、PAKが癌化においてAktと関連する可能性が考えられる。我々はまたAktが生存を促進するメカニズムについて検討しており、これまでにBadならびにNur77がAktによりリン酸化を受けると14-3-3タンパク質と結合することを報告した。本研究において、生存シグナルによってAktが活性化していても、細胞ストレスにより活性化したJNKが14-3-3を直接リン酸化し、このリン酸化によって14-3-3と結合していたアポトーシス誘導分子が解離して細胞死を引き起こす、というメカニズムを明らかにした。この結果は、哺乳類細胞における死誘導シグナルと生存促進シグナルの拮抗作用の分子機構の一部を明らかにしたものと考えている。
