二量体化ライブラリーを用いた増殖・抗アポプトーシス関連遺伝子の単離

研究概要	本研究ではDNA gyraseあるいはエストロジェンレセプターなどの人為的2量体化システムを用いてライブラリーを作成し、細胞増殖あるいは抗アポプトーシス活性を指標に、2量体化によって活性化を受ける新規分子の単離を目標とした。当初の計画どうりレトロウイルスライブラリーを用いることとし、エストロジェンレセプター、DNA gyraseそれぞれと融合蛋白質となるようなライブラリーを作成し、マウスIL-3依存性株であるBa/F3細胞に導入した。その後IL-3非依存性に増殖するクローン、ガンマ線照射後も生存するクローンを単離し、genomic PCRによりレトロウイルスにより染色体中に導入された遺伝子を単離した。その結果、IL-3非依存的に増殖する株からは融合蛋白質をコードすると思われる配列が見いだされず、IL-3非依存性の増殖はインサーションミュタジェネシスによるものと予想された。一方、ガンマ線照射に対し耐性になる株からはヒストンアセチレース2のマウスホモログが単離された。このものが、ガンマ線照射に対して実際に耐性となるのかどうか検討を加えている。
