癌転移におけるαvβ3インテグリン・結合分子オステオポンチンの機能解析

研究概要

本年度においては癌細胞において発現し、その転移性との関連が推測されている細胞接着分子、オステオポンチンについて、癌細胞転移におけるこの分子のホスト側についての役割を検討した。癌細胞としては、マウスの悪性黒色種B16をもちいた。まず、この細胞の接着性のオステオポンチン依存性を、インビトロにおいて明らかにした。この上で、B16細胞を左心室内より注入することにより、実験的な転移モデルを作成して、オステオポンチンノックアウトマウスと野生型マウスについての転移の効率を検討した。
動脈内注入モデルでは以前の報告通り野生型マウスに骨に対して転移巣(11.5+/-2.5,n=18)が観察された。これに対してオステオポンチンノックアウトマウスにおいては骨で約50%(5.4+/-1.7,n=19,p<0.05)の有意な転移効率の低下が見られた。副腎でも野生型マウスに対してオステオポンチンノックアウトマウスにおいては有意な転移効率の低下(17.8+/-5.5,n=18対6.5+/-2.8,n=19,p=0.0514)が観察された。静脈内注入モデルで野生型マウスで肺に対して多数の転移巣(126.7+/-42.1,n=11)が観察された。これに対してオステオポンチンノックアウトマウスにおいては約3分の1(37.8+/-11.4,n=17,p<0.05)の有意な転移効率の低下が見られた。本研究の結果、ホスト側のオステオポンチンの存在は、B16悪性黒色腫の転移を促進する活性を持つことが明らかとなった。
