造血幹細胞の増殖・分化とその異常に関する研究

研究概要	マウス胎生期における造血幹細胞の発生とその異常を明らかにするとともに、様々な方法を用いてヒト造血幹細胞の性状、増殖・分化過程を明らかにすることを目的として研究が行われ、以下の研究成果が得られた。貧血のため胎生致死となるEPOR^<-/+>マウスとhGM-CSFR-Tgマウスを交配したマウス胎児肝細胞をGM-CSFで刺激すると多数の赤芽球コロニーが無血清下で形成された。赤芽球産生において必須と考えられていたEPORシグナルはhGM-CSFRシグナルによって完全に代用できることが明らかとなった。サイトカインの造血における主たる作用は細胞の増殖と生存であり、分化の特異性は極めて低いと考えられた。造血幹細胞の発生する胎生10.5日のAGM領域から造血幹細胞自己複製支持能を持つストローマ細胞株を樹立した。現在までにこのストローマ細胞株から20を超える新規遺伝子が同定され機能解析が進行中である。現在その内の1つのリコンビナント蛋白に対する抗体が作成され、造血幹細胞のある分画を認識している可能性を示すデータが得られつつある。ここ研究は造血幹細胞の生物学的研究にとって有用であるばかりでなく、臨床応用される可能性も示している。 Wild typeおよびKostmann症候群患者で見られるG-CSFR遺伝子異常と同様の2種類の遺伝子異常(マウス718,731)を持つTgマウスを作成した。C末端を欠失した2種類の変異G-CSFR-Tgマウスでは共に好中球減少が見られ、本症候群のモデルとなることが明らかとなった。wild typeおよび2種類の変異G-CSFR-TgマウスにG-CSFを1年間投与した。変異G-CSFR-TgマウスへG-CSFの投与により、3血球系とも著明に増加した状態が続いているが、今のところ白血病の発生は見られていない。現在、2^<nd>ヒットとなる刺激を加え、白血病発症の有無を検討中である。
