受容体からの分化と腫瘍化のシグナル

研究概要

我々は、既知の抗アポトーシス分子とは全くホモロジーを有さない新規抗アポトーシス分子Anamorsinを同定した。Anamorsinの発現は、サイトカイン刺激により誘導され、Anamorsinを遺伝子導入した細胞はサイトカイン除去により引き起こされるアポトーシスに抵抗性を示した。
Anamorsin^<-/->マウスは肝臓、脾臓の造血組織が萎縮し、著明な貧血により胎生14.5日以降に死亡した。Anamorsin^<-/->胎児肝組織では赤芽球のアポトーシスが認められ、成熟した赤芽球が減少していた。Anamorsin^<-/->胎児肝においては、造血幹/前駆細胞分画の絶対数は減少していなかったが、各種サイトカイン存在下にin vitroコロニーアッセイを行うと、Anamorsin^<-/->胎児肝細胞の造血幹/前駆細胞のコロニー形成能は著減し、特に赤血球系コロニーの形成は全く認められなった。以上の結果より、Anamorsin^<-/->マウスでは、胎児肝にある造血細胞のアポトーシス亢進と分化障害により、二次造血が障害され造血不全をきたすことが明らかとなった。
さらに、各種造血器腫瘍細胞におけるAnamorsinの発現をmRNA定量法により検討した。マントル細胞リンパ腫など一部造血器腫瘍細胞においてAnamorsinの高発現が認められ、Anamorsinが造血細胞の腫瘍化に関与している可能性が考えられた。
