研究概要	1.PolII転写開始におけるヒトTFIIEβの二本鎖DNA結合領域の機能解析ヒトTFIIEは、αとβがα2β2の4量体を形成し、転写開始と伸長への移行に機能する。転写開始では、TFIIEがプロモーターの転写開始点すぐ上流に結合し、複合体活性化とDNAの一本鎖への開裂を助ける。昨年度、TFIIEβの中央コア領域が二本鎖DNAとの結合領域で、羽状-ヘリックス構造をとることを明らかにした。今年度、この領域のDNA結合アミノ酸残基の機能解析を行った。DNA結合残基に点突然変異を導入したところ、転写活性が著しく低下した。また、この領域が転写開始の際、DNAとの結合に重要であることを初めて明らかに出来た。 2.線虫TFIIEを用いたPolIIの転写開始から伸長への移行段階の解析線虫TFIIEは、アミノ酸配列をヒトと比較するとαで29%、βで40%一致していた。今年度、ヒトと線虫TFIIEのサブユニットを組み合わせたキメラTFIIEを大腸菌にて発現・精製し、ヒト転写系にてヒトTFIIEと機能互換性を検討した。ヒトαと線虫βのTFIIEは、直鎖状鋳型を用いたヒト転写再構成系で解析すると、転写開始はヒトTFIIEの50%程度の活性を示したが、伸長への移行は5%以下に低下していた。この結果より、TFIIEは伸長への移行段階に機能することが示された。この段階で、PolIIは最大サブユニットのC末端の7アミノ酸繰り返しCTD配列の2番目と5番目のセリンをTFIIHがリン酸化することで活性化され、その際TFIIEがTFIIHのキナーゼ活性を促進して制御している。上記キメラTFIIEを用いてPolIIのCTDリン酸化を調べると、CTD5番目のセリンリン酸化促進能が著しく低下していた。以上の結果から、CTDの5番目セリンリン酸化と転写伸長への移行活性は密接な関連性があることが示唆された。
