研究概要

上皮細胞系の増殖、分化の制御機構は、ヒトがん発生のメカニズムを知る上で必須といえる。本研究では重層偏平上皮を構成するケラチノサイトを用いて、上皮細胞の増殖・分化の制御機構を明らかにすることを目的とした。
特に扁平上皮組織の終末分化におけるシグナル伝達経路をCキナーゼη分子種(PKCeta)を中心に解析した。
1)終末分化に伴うG1期停止は、PKCetaがcdk2/cyclin E/p21複合体と結合し、cdk2活性を抑制することによる。このPKCeta/cdk2/cyclin E/p21の結合に必要なアダプター分子として、14-3-3sigmaを同定した。14-3-3sigmaは、PKCetaと共に細胞老化も制御していた。老化誘導には14-3-3sigmaの局在変化が関与していることが示唆された。
2)p21の146Serine残基がPKCetaによりリン酸化され、p21の局在に影響を及ぼしていた。
3)PKCetaトランスジェニックマウスは、上皮組織の分化高進所見を示す。さらにE-カドヘリンの局在異常を含む細胞間接着や細胞極性の異常を見いだした。
4)新規PKCとして、PKCthetaのalternative splicing variant,PKCtheta IIをクローニングした。PKCtheta IIは精巣の発生段階に依存して、精細管に特異的に発現している。
