胚性腫瘍細胞の分化機構とヒストンアセチル化

研究概要	胚性腫瘍細胞F9のレチノイン酸やアデノウイルスE1Aによる分化誘導は、c-junのトランス活性化によって引き金が引かれる。c-junのプロモーター上にこのトランス活性化に必要なエレメントDREを同定し、ここに転写因子複合体DRFが結合することを明らかにしてきた。DRF複合体はコアクチベーターp300,転写因子ATF-2,JDP-2及びJunDより構成される。転写因子ATF-2は、分化誘導に伴い、N末端がリン酸化される。また、このリン酸化はJNKによって駆動され、このリン酸化によってATF-2はヒストンをアセチル化する活性が促進されることを初めて見い出した。ATF-2は従来の転写因子とは異なり、ヒストンH2B,ヒストンH4をアセチル化するヒストンアセチル化酵素である。また、このATF-2をベートとして酵母ツーハイブリット法を行い、新規の転写因子JDP-2を同定した。JDP-2はATF-2をはじめとするbZipモチーフを有する転写因子とヘテロ二量体を形成し、c-junの転写活性を抑制する。未分化状態ではJDP-2はDRFに結合し、c-junを活性化しているが、分化に伴いJDP-2のDNA結合能は消失することが引き金を引くと推定される。アセチル化と脱アセチル化との関係を現在検討中である。
