マウス大腸癌転移モデルに基づくヒト大腸癌転移の遺伝子診断法の研究

研究概要	平成12年度には、宿主との相互作用を考えるために有用である同系マウスを用いた大腸癌肝転移モデルを作成し、肝臓に高転移性のバリアント細胞(colon38-SL4細胞)を選別し、それ由来のクローン細胞を得た。これらの細胞に特徴的な形質は、肺転移性の高い細胞の形質とは異なり、増殖性が高い一方、運動性、接着性、マトリクス分解酵素のレベルに関しては、親株と大きな差異がなかった。ディファレンシャルディスプレイ法及びアレイ解析法によって、この高転移バリアント細胞のにおいて発現量の変化している複数の遺伝子を同定することに成功した。発現レベルの差異は、ノザン解析及び半定量RT-PCR法によって確認した。これらの遺伝子は、これまで肺に高転移性の癌細胞の解析から転移に関係するとされて来た細胞形質とは全く異なる分子をコードしているものであった。これらの複数の遺伝子の発現が、同時に共通の機構で制御されていると言う証拠は得られなかった。また転移性との直接の相関も不明であった。これら遺伝子の内で、in vivoで増殖している細胞においても発現レベルの違いが検出できるものを選んで、ヒト相同遺伝子検出のプローブをデザインし、ヒト大腸癌及びその肝転移巣における発現レベルを比較した。転移性と相関して発現する複数の遺伝子の中で、神経系に発現して細胞の移動に関係する細胞外分子であるreelinは、転移巣で発現が相対的に高い傾向にあった。マウス大腸癌細胞においてこの分子の発現レベルを遺伝子導入によって亢進させると、細胞の足場非依存的な増殖性が上昇することを証明した。
