マウスモデルに基づくヒト大腸癌転移形質のバイオインフォーマティックスによる解析

研究概要

本研究は、同種同所移植によって肝転移を形成しやすいマウス大腸癌バリアント細胞SL4とその親株colon38細胞を用いて、In vitro及びin vivoで増殖させた際の遺伝子、蛋白質、細胞表面糖鎖の発現を、遺伝子発現解析、プロテオミクス解析、糖鎖インフォーマティックス解析によって比較検討し、それらの相互関係及びこれらの癌細胞の生物学的な性質との関連を明らかにすることを目的とした。これらの知見をヒト大腸癌の個性の診断に役立てることを目指している。単独で腫瘍の生物学的な形質との相関が見られた遺伝子、蛋白質及び糖鎖に関しては、細胞株において強制発現または細胞の選別とクローニングによって、生物学的な形質、特に肝転移性に変化が見られるかどうかを解析することも射程に入れている。これまでに、マウス大腸癌colon38細胞より肝臓に高転移性を有する細胞株(SL4)及びそれに由来する高移性クローンを得た。SL4細胞における最も顕著な細胞形質の違いは増殖速度の違いであり、従来肺への転移性に相関すると言われていた、細胞外マトリクスの分解活性、接着性、運動性、浸潤性などには全く差異が無かった。本年度はこれらの細胞の遺伝子発現プロファイルを解析し、低転移性細胞または高転移性細胞に発現の高い複数の遺伝子を既に同定した結果肝臓に高転移性を持つ細胞ではEphB2/Nukの発現が低いことから、消化管の上皮細胞の特徴とも考えられる「環境による増殖性制御機構」が破綻しているという可能性を検証し、β-カテニンシグナルの低下及びTGF-βシグナルの低下という現象を見い出した。またこの細胞が微小肝転移を形成してから肝転移を確立するまでの間に、ガラクトース型レクチン発現マクロファージと混在する時期があるが、このレクチンと癌細胞の相互作用が、転移確立との間に直接因果関係を持つものでないことを検証した。
