薬剤感受性と血管新生の分子標的から見たがんの診断と治療

研究概要

1.薬剤感受性を制御する分子標的としてのABCトランスポーターを中心に検討した。その結果、(1)MRP3が胆汁酸で発現上昇し、プロモーター上のFTF結合サイトが重要であることを示している。腸肝循環を助けるMRP3の新しい役割について提示した。さらに、MRP3がその排出に関与するであろう抗がん剤としてエトポシドやシスプラチンを報告した。
(2)MDR1,MDR3,MRP1,MRP2,MRP3のうち、MRP2がC型肝炎ウイルスの患者の肝細胞で有為に減少していることとさらに再発膀胱がんMDR1とMRP2が有為に上昇していた。
2.がん血管新生を制御する分子標的に関してはTP/PD-ECGF, VEGF受容体、ならびにEGF受容体を中心に検討した。その結果、(1)IFNγによるTPの発現上昇にはGASエレメントによる転写制御が関与していた。
(2)SU5416による血管新生阻害には、VEGF受容体のうちKDR/Flk-1だけでなく、Flt-1の機能阻害が関与することを示唆した。
(3)EGF受容体を標的とする分子標的薬剤ZD1839の抗腫瘍効果には血管新生の阻害が重要であることを示唆した。
