研究概要

がんの「抗体療法」の視点から基礎研究成果を臨床へ迅速に引き上げていくtranslation researchの実践を、以下の項目から検討してきた。B型肝炎ウイルスのなかで肝癌発症リスクがgenotype間で異なることを見出したため、ハイリスクgenotypeの迅速血清診断法としてモノクローナル抗体を作製、臨床応用への可能性を検証している。ガングリオシドGD2抗原が肺小細胞癌に高発現し増殖亢進作用をもたらし、GD2抗体によるJNK系を介したアポトーシス誘導増殖抑制効果、抗癌剤併用効果、さらにヒト化抗体、IL2融合抗体のより強力な腫瘍抑制効果をin vitro及びin vivo(マウス)で確認しえた。多発性骨髄腫関連原癌遺伝子MUM1/IRF4が、慢性リンパ性白血病での予後不良因子、骨髄腫の成立・進展に関わる分子であることを明らかとした。またMUM1関連分子候補をマイクロアレイにて4つ同定し、そのうちのひとつMIG遺伝子はMUM1によって誘導されることを確認した。抗変異EGFレセプター抗体(3C10)では脳内腫瘍移植マウスの実験で優れた腫瘍への集積と99mTc標識が可能であることを示した。細胞の遊走、細胞間接着に関わるケモカインCCR4が成人T細胞性白血病(ATL)で高発現し、予後不良因子、皮膚浸潤との関連を見出した。さらにATL及びCCR4発現T細胞系腫瘍細胞株でキメラCCR4抗体によるADCC活性と増殖抑制効果を確認できた。抗体を用いた前臨床試験への導入をにらみ、さらなる検討を加えていく。
