温熱療法における分子シャペロン・遺伝子治療の応用の基礎的研究

研究概要	【目的】本研究では、変性した蛋白質構造を正常な蛋白質構造に戻す働きがあるとされる分子シャペロン(グリセロール)を用い、変異型p53蛋白質に正常型p53蛋白質の正常機能を回復させることにより、効率のよい温熱および放射線、制癌剤による癌治療の開発を目指した。【方法】p53欠損ヒト肺癌細胞H1299に変異部位の異なる変異型p53遺伝子(codon175,Arg→His;codon248,Arg→Trp;codon273,Arg→His)を導入した細胞(H1299/mp53)、p53の表現型が正常型の舌扁平上皮癌細胞SASに変異型p53遺伝子(codon248,Arg→Trp)を導入した細胞(SAS/Trp248)および変異型p53を保有する甲状腺未分化癌細胞8305c(codon273,Arg→Trp)とヒト口腔扁平上皮癌細胞株Ca9-22(codon248,Arg→Trp)を用いた。グリセロール処理は放射線あるいはCDDP処理の2日前に培地に添加した(最終濃度0.6M)。コロニー形成法で感受性を測定し、ヘキスト染色法及びDNA ladder検出法でアポトーシスの解析を行った。SAS/neo細胞、SAS/Trp248細胞または8305c細胞をヌードマウス(Balb/cAJcl-nl)の大腿皮下に移植し、1.2Mグリセロールを3日間連続局所投与および2日目のグリセロール局所投与直後にCDDPを腹腔内投与あるいはX線を腫瘍部位に照射した。【結果】グリセロールによりH1299/mp53、SAS/Trp248、8305cおよびCa9-22細胞のX線感受性が増感し、アポトーシスが増強された。グリセロール処理をした8305cあるいはSAS/Trp248細胞では、CDDPによるアポトーシス誘導の増強が見られ、これら細胞のヌードマウス移植系においてはX線あるいはCDDPによる腫瘍増殖抑制がグリセロール処理で増強された。温熱の場合はグリセロールによる感受性の増強効果は見られなかった。このシャペロン効果はp53遺伝子の変異部位に依存しないことや正常型p53蛋白質の機能亢進によるものではないことを今年度新たに確認した。【考察】グリセロールを介した変異型p53蛋白質の活性回復により、X線あるいはCDDP処理後にp53依存的アポトーシスが誘導され、感受性あるいは増殖抑制が増強されることが示唆された。【結論】変異型p53を保有する癌患者に対しては分子シャペロン治療を併用した放射線あるいは抗癌剤治療が有効であることが培養細胞系およびヌードマウス移植系で強く示唆された。
