分子シャペロンを併用した放射線・温熱癌治療法の基礎的研究

研究概要

本研究では分子シャペロン効果を臨床応用により近づけるために、癌抑制遺伝子p53のみ異なった培養細胞系およびそれら癌細胞をヌードマウスへ移植した実験腫瘍モデル系で、分子シャペロン治療と放射線・温熱治療の併用がより効果の高い新しい治療法となり得るかを検討した。その結果、以下のような結果を得た。
1.p53欠損ヒト肺癌細胞(H1299)に正常型p53遺伝子(H1299/wtp53細胞)、変異型p53遺伝子(codon 248,Arg→Trp)を導入した(H1299/248細胞)。H1299/248細胞を前もってグリセロール処理(0.6M)すると、X線感受性が増強された。さらに、ヘキスト染色及びDNA ladder検出法によりアポトーシスの解析を行ったところ、X線あるいはグリセロール単独ではアポトーシスがほとんど誘導されなかったが、グリセロール処理とX線照射を併用するとアポトーシスが増強された。H1299/Wtp53およびH1299/neo細胞ではこのような増強効果は見られなかった。
2.H1299/248細胞をグリセロール処理すると変異型p53蛋白質がX線照射によってDNA結合能を回復するかどうかをゲルシフト法で調べたところ、bax遺伝子のプロモーター領域に変異型p53蛋白質が結合できることを確認した。
3.グリセロールの分子シャペロン効果が移植癌でも見られるかどうかを検討するため、mp53導入舌扁平上皮癌細胞SAS/Trp248細胞(mp53)またはSAS/neo細胞(wtp53)をヌードマウス(Balb/cAJcl-nl)の大腿皮下に移植し、グリセロール局所投与後にマウス体重1mgあたり6μgのCDDPを腹腔内投与した。経日的に腫瘍径を計測し、腫瘍重量を算出した。その結果、グリセロールをあらかじめ投与すると、mp53腫瘍においてもwtp53腫瘍と同様にCDDPの抗腫瘍効果を得た。
4.グリセロールが変異型p53腫瘍の放射線感受性も高められるか、またp53遺伝子の突然変異部位が異なる癌細胞の移植腫瘍でもグリセロールが分子シャペロン効果を示すかについて検討した。変異型p53遺伝子(codon 273,C:G→T:A)を有する8305c細胞でもグリセロール処理により放射線照射群の相対腫瘍成長遅延時間は延長することが分かった。p53遺伝子の突然変異部位が異なる移植腫瘍においてもグリセロールは放射線感受性を増強することが示された。さらに、アポトーシス関連蛋白質のactive caspase-3、85kD PARPの発現を免疫染色にて確認した結果、グリセロール、CDDP併用処理群で陽性細胞の増強を認め、アポトーシスが誘導されていることが確認された。
以上の実験結果より、変異型p53蛋白質を標的としたグリセロールによる分子シャペロン治療の有効性が培養細胞系およびヌードマウス移植系で強く示唆された。
