腫瘍血管新生におけるエリスロポイエチンの役割の解明

研究概要

Erythropoietin(Epo)は低酸素により誘導され、赤芽球の分化に関与し、赤血球の産生を増加させる。近年、Epoが血管新生に関与するという報告が見られる。我々はラット化学物質誘発肝癌モデルを用いてEpo,Epo-r,VEGF,flt(VEGF receptor)などの免疫組織化学染色による形態学的解析を行うとともに、それらの蛋白量を定量した。本実験の結果、肝腫瘍からEpo-rが検出されたが、正常肝組織、肝硬変組織からは検出されなかった。また、腫瘍内Epo濃度と血管密度は相関した。肝腫瘍の血管新生にEpoが関与しているものと推定された。
また、ヌードマウスにヒト子宮癌および卵巣癌細胞を移植し、抗Epo抗体を局所に投与した。その結果、腫瘍容積は縮小し、免疫組織染色を行ったところ、腫瘍細胞のアポトーシスが確認された。腫瘍の縮小は抗Epo抗体の容量依存的におこり、無治療の移植腫瘍ではEpo,Epo-rのmRNAおよびEpo蛋白が発現していた。悪性腫瘍の進展にEpoが関与し、このシグナルを阻害することが治療効果に結びつくことを明らかにした。
