癌および正常細胞の抗癌剤感受性を規定するサプレッサー型耐性因子の同定

研究概要	本研究では、遺伝子の機能不全あるいは発現低下による耐性化に関与する遺伝子の同定を試みた。マウスNIH3T3細胞のcDNAをLNCXレトロウイルスベクターに組み込んだライブラリーをNIH3T3細胞に導入し、種々の抗癌剤で選択して耐性クローンを得た。得られた抗癌剤耐性クローンに組み込まれたプロウイルスに挿入されていたcDNAを解析した結果、NADH-ubiquinone oxidoreductaseのchain5のアンチセンスを発現する細胞がAdriamycin耐性を示すこと、vacuolar assembly protein VPS41の部分ペプチドを発現する細胞がpaclitaxel耐性を示すことなどが明らかになった。また、Adriamycin耐性クローンAc2株は1.2kbの新規遺伝子断片を組み込んでいた。この遺伝子をTASPと名付けた。ヒト、マウスのTASP全長cDNAがコードする蛋白はともに450アミノ酸よりなり、細胞膜を7回貫通する構造をもつG-protein受容体と推定された。TASPの発現は調べた全組織で見られたが、とりわけtestisで高い発現を示した。Ac2株に組み込まれていたretrovirusはTASPの3'側のantisenseを発現するものであったが、このantisense発現retrovirusを新たに構築してNIH3T3細胞に導入してG418で選択したところ、遺伝子導入細胞はAdriamycinに1.5倍の耐性を獲得した。さらに、ヒトTASPの全長cDNAをpHa-IRES-DHFRベクターに組み込み、NIH3T3細胞に導入後、methotrexateで選択して遺伝子導入細胞を得た。遺伝子導入細胞で50-kilodaltonのTASP蛋白の発現が見られ、この細胞はAdriamycinに1.4倍高感受性であった。以上より本研究で単離された新規遺伝子TASPは細胞のAdriamycin感受性に関与していると推定された。
