研究概要

平成13年度がん特定領域研究補助金によって以下の成果を得た。
1.がん抑制遺伝子p53は、CDK inhibitorであるp21WAF1の誘導を介して細胞周期をG1期で停止させる。そこで、p21WAF1プロモーターを活性化する低分子化合物はp53欠損がん細胞に対する化学療法剤になる可能性がある。そこで探索系を構築し、その探索を行った結果、土壌より分離したカビFusarium sp. RK97-94より、P53欠損細胞の増殖を阻害することが期待されるp21WAF1活性化物質Lucilactaeneを発見した。作用を追究している。
2.乳癌細胞はPTHrP(副甲状腺ホルモン類縁タンパク)を分泌し、破骨細胞を活性化し骨破壊を促進し、骨転移することが明らかにされている。乳癌の骨転移の阻害物質として、PTHrPによる破骨細胞分化を阻害する物質を微生物生産物中に探索し、Cunninghamella sp.の生産物中より新規構造を有するf8756を見い出した。作用機構ならびに骨転移に対する効果を検討し、有用性を追究する。
3.がん細胞の増殖に間質細胞が関わっていることが明らかにされつつある。前立腺がん細胞と間質細胞との共培養系を用いてがん細胞の増殖を阻害する物質を探索し、Phthoxazolinにその作用を見い出した。移植腫瘍を用いて制癌作用を検討する。
4.ヒト前立腺がん細胞の中でDU145細胞の培養上清が前立腺間質細胞のアンドロゲンレセプターを減弱させることを見い出した。前立腺がんのアンドロゲン非依存的な増殖に関与する可能性が示唆され、この結果を基に、前立腺がんの悪性化を阻害する新たな探索系を構築する。
