ビタミンE転送蛋白質遺伝子と発ガンリスクの個体差に関する分子疫学的研究

研究概要	ビタミンE転送蛋白質(α-Tocopherol Transfer Protein;α-TTP)遺伝子の変異や多型と、血中ビタミンE濃度およびがんを含めた疾患発症との間の相関を調べることを目的として、まずヒトα-TTP遺伝子多型の検索を行った。まず、ヒトα-TTP遺伝子の全長を単離して、その構造を決定した。次いで、24名の日本人を対象としてα-TTP遺伝子の5個のエクソンとその周辺のイントロン領域、及びプロモーター領域に対してPCRプライマーを設定してPCR-SSCP法による遺伝子多型のスクリーニングを行った。その結果、互いに強い連鎖不平衡にある2つの頻度の高い遺伝子多型(プロモーター領域多型とイントロンDのC/T多型)を同定した。特に、プロモーター領域の繰り返し多型[-335A(n)多型]については、DNAの塩基配列を調べた結果、ヒトα-TTP遺伝子の転写開始地点より数えて335塩基5′上流地点に、アデニンが10回繰り返すものが最も高頻度に見られたが、その他にもアデニンの繰り返しが、11回、9回、12回繰り返すものを認めた。このプロモーター領域多型は新規の遺伝子多型であり、この多型に関しては培養細胞における感染発現実験を通じて、これがα-TTP遺伝子の転写活性に影響を与える機能的な多型であることを確認した。遺伝子型の決定が容易なイントロンD多型と血中ビタミンE濃度との相関を150名のボランティアに関して調べたところ、C/C,C/T,T/Tの順に血中ビタミンE濃度が低下する傾向が見られた。これらの遺伝子多型とがんを含む疾患発症との間の相関について現在検討中である。
