蛋白質リン酸化・脱リン酸化反応を介する細胞増殖と細胞癌化の制御

研究概要	がんの発症進展に関わるがん原遺伝子やがん抑制遺伝子は、もともと細胞の増殖・分化を厳密かつ巧妙に制御している。本研究では、哺乳類細胞の増殖・分化の基盤となっている増殖シグナル伝達機構や細胞周期の調節に焦点をあて、細胞膜からのチロシンキナーゼ活性を基点とする細胞の増殖・分化シグナルが細胞周期制御のシグナルに変換されていく分子機構の解明を試みた。また、G2-M期の制御機構をM期キナーゼによる蛋白質リン酸化反応に着目して解析した。具体的な研究成果は以下の通りである。(1)Lynチロシンキナーゼの標的として見出したBANKは、チロシンリン酸化されるとIP_3受容体と会合し細胞内Ca^<2+>動員を増強させる作用を持つ新規scaffold蛋白質である。このBANKによるIP_3受容体活性化経路はPLCγの活性とは独立した新規シグナル系である。(2)Ki-1リンパ腫の原因となる活性型ALKチロシンキナーゼNPM-ALKはSNT2アダプター蛋白質と会合し、その会合が細胞がん化において重要である。(3)癌抑制遺伝子産物TDbはRas/MAPキナーゼ依存的な細胞増殖を抑制するが、v-FosやLarge T等による増殖シグナルは抑制しない。TQbはHDACと会合してcychin D1遺伝子の転写を抑制する。その結果Rb蛋白質のリン酸化が抑制され、細胞周期進行が阻害される。Erk1/2によってリン酸化されたTobはcyclin D1の発現は抑制しない。また、tobとp53の二重欠損マウスでは加算的にがん発症が見られた。更に多くのヒト肺がんでTobの発現低下が見られた。(4)ハエがん抑制遺伝子産物のホモログである2種のキナーゼLATS2とLATS1は中心体や紡錘体に局在している。LATS2はLATS1との会合を通してBcl2をリン酸化し、微小管を標的とする抗がん剤taxo1による細胞死誘導に関わる。LATS2はM期チェックポイント蛋白質とし機能していることが示唆された。
