研究概要	細胞周期の制御機構は、がん化や老化を含めた細胞増殖を基盤とする生命現象にかかわり、その特異的阻害剤は基礎/応用両面から重要である。細胞周期のG1,G2停止を誘導するトリコスタチンA(TSA)、トラポキシン(TPX)の作用機構を解析し、ヒストンデアセチラーゼ(HDAC)を阻害することを明らかにし、HDACが新しいがん化学療法の分子標的として重要であると考え、新しいHDAC阻害剤の開発を行った。HDACとTSAの共結晶構造の情報からTSAの持つヒドロキサム酸が酵素活性中心の亜鉛とキレートすること、また、TPXのエポキシケトンが酵素との共有結合に必要であることが示唆された。TSA,TPXは生体内では不安定なため、トラポキシン類似の種々の環状テトラペプチド骨格にTSAの活性基ヒドロキサム酸を導入したハイブリッド化合物CHAPを作成した。CHAPの活性はヒドロキサム酸の存在とヒドロキサム酸までの側鎖の長さに依存していた。一群のCHAP化合物は、、親化合物であるTSAやTPXを越える阻害性を示した。さらにCHAP化合物はHDAC6よりもトHDAC1に対してより強力に阻害することが判明し、TSAよりも大きなキャップ構造を持つことが酵素選択性に関わっていることが示唆された。主としてG1期停止を話導するラディシコール(RAD)の作用機構を解析した。ビオチン化したRAD誘導体を用いててRADはHsp90とATP-クエン酸リアーゼの双方に結合して阻害することを明らかにした。RADのオキシム体は動物実験で強い抗腫瘍活性が認められており、これらの結合タンパク質の機能を阻害することが原因と考えられた。
