研究概要

Wntシグナル伝達経路の異常においては、β-カテニンが安定化してTcfが活性化されることが知られている。これまでに、私共はAxinに種々の蛋白質が結合することにより、β-カテニンのリン酸化とユビキチン化が促進されβ-カテニンが分解することおよびTcfの活性にSUMO化が重要な役割を果すことを明らかにしてきた。今年度はβ-カテニンの安定性の制御について次の点を明らかにした。
(1)ダイナミンK44Aはアドレナリン受容体刺激によるエンドサイトーシスを抑制することが知られているが、ダイナミンK44AはWnt-3a依存性のβ-カテニンの蓄積を抑制した。また、EpsinのENTH領域がインスリンとEGFの受容体のエンドサイトーシスを阻害するが、ENTH領域もWnt-3a依存性のβ-カテニンの蓄積を抑制した。さらに、クラスリンを安定化させ、受容体のエンドサイトーシスを阻害するダンシルカダベリンを細胞に作用させると、Wnt-3a依存性のβ-カテニンの蓄積が減少した。したがって、Wnt-3aによるβ-カテニン経路の活性化には、受容体のエンドサイトーシスが必要ということが示唆された。
(2)Aキナーゼ(PKA)はプレセニリンと結合することにより、β-カテニンを分解することが報告されているが、PKAを活性化するとβ-カテニンは安定化した。TNF依存性のIκBの分解には影響が無いことから、PKAの作用はβ-カテニンに特異的であると考えられた。PKAはAxin複合体においてAPCをリン酸化して、APCとAxinの結合を阻害した。さらに、PKAはβ-カテニンのユビキチン化を抑制した。これらの結果から、PKAはAxin複合体の機能を抑制することにより、β-カテニンを安定化させると考えられた。
